Once you realize you have a reduced CYP2D6 enzyme, what, if any, drugs do you need to compensate for this deficit; and/or, what natural or form of supplements, and allowed foods, do you need to consume to increase the enzyme activity and effectively metabolize appropriately- without causing a toxic build-up and further depletion to your skeletal structure?

I took it for 3 months for surgery 3.5 years ago and then I haven taken it for 2+ months now, starting in Sept. 2019.

Since my first surgery, I have continued to feel more depressed that I ever was before surgery. Now, even 1/4 of a 25 mg pill make me feel better but after 2-3 days the aching comes back!

This article was very interesting but I’m not sure where to go from here. Could Tramadol have LONG-TERM impacts on mood and cognitive functioning? Thanks

Makes me think that in addition to affecting how anti-depressants are processed that it could be a contributor to her depression. Can you share additional studies/data on this? My daughter is a 17 year old Caucasian, this seems to be a rare condition for north american Caucasians, so there is not enough data out there. I am looking to anything to help her live her best life. Your response is appreciated.

My 8 year old had a surgery at about 2.5 years old. Before surgery he was healthy and all milestones normal walking and talking. During the surgery they had problems keeping him under, the surgery was done and he was not the same since. In the next two weeks his speech slowed until it stopped completely and he became very wobbly. Falling all the time. Doctors kept telling me he was in shock and to just love on him. Weeks went by doctor visit after doctor visit. They tried to tell me my son was deaf, autistic, Asperger’s, some forms of down or anything to make us go away. In an ENT appointment I broke down sobbing when I told them this happened in a matter of days of a surgery and how my son gets sick after Tylenol , hives with antibiotics , Benadryl put him to sleep for a day. The doctors eyes lit up and she ordered a pharmacogenetic test immediately. She also advised to stop any medication and supplements. They also tested for other things. The results came back the geneticist called me, advised to not give him any medications and come in asap. The only thing that came back was that his CYP2D6 gene wasn’t right. She said in that gene 1 is paralyzed, 1 works negatively, and an extra that works fine…

So here we are 6 years later, my son has had lots of therapy and his talking is so much better. I just have no answers about what else to watch out for and if he should have a certain diet. Nothing, the geneticist said to me she wished she never tested him because she has no answers for me. I don’t know what to do. Can you help me Please?

Years ago, I took Paxil (Paroxetene) for 2 days, then stopped. I then began to have muscle twitching that made it nearly impossible to sleep for up to one year, and that came intermittently for 6 years following.

Years later, I had genetic testing done, and found that, indeed, I have two defective alleles of CYP2D6.

I have no doubt that my poor CYP2D6 metabolism was the cause of my adverse reaction to Paxil, which no doctor at the time could understand.

What I am likewise amazed by, however, reading your article, is the further correlation to personality traits.

I totally identify with these:

– increased anxiety (was the reason why I was prescribed Paxil in the first place)
– higher impulsivity
– performing well in tasks that demand sustained attention – have always been able to focus for long hours
– “high conscientiousness/responsibility, orderliness, and the pursuit of achievement through perseverance.”

Also, you mention a correlation to autoimmune disease. Indeed, I was diagnosed with Hashimoto’s hypothyroidism at the age of 12.
